    A V-Diagram for the Design of Integrated Health Management for Unmanned Aerial Systems

    Designing Integrated Vehicle Health Management (IVHM) for Unmanned Aerial Systems (UAS) is inherently complex. UAS are a system of systems (SoS) and IVHM is a product-service, thus the designer has to take into account many factors, such as: the design of the other systems of the UAS (e.g. engines, structure, communications), the split of functions between elements of the UAS, the intended operation/mission of the UAS, the cost verses benefit of monitoring a system/component/part, different techniques for monitoring the health of the UAS, optimizing the health of the fleet and not just the individual UAS, amongst others. The design of IVHM cannot sit alongside, or after, the design of UAS, but itself be integrated into the overall design to maximize IVHM’s potential. Many different methods exist to help design complex products and manage the process. One method used is the V-diagram which is based on three concepts: decomposition & definition; integration & testing; and verification & validation. This paper adapts the V-diagram so that it can be used for designing IVHM for UAS. The adapted v-diagram splits into different tracks for the different system elements of the UAS and responses to health states (decomposition and definition). These tracks are then combined into an overall IVHM provision for the UAS (integration and testing), which can be verified and validated. The stages of the adapted V-diagram can easily be aligned with the stages of the V-diagram being used to design the UAS bringing the design of the IVHM in step with the overall design process. The adapted V-diagram also allows the design IVHM for a UAS to be broken down in to smaller tasks which can be assigned to people/teams with the relevant competencies. The adapted V-diagram could also be used to design IVHM for other SoS and other vehicles or products

    This is the second half of a two paper series covering aspects of the no fault found (NFF) phenomenon, which is highly challenging and is becoming even more important due to increasing complexity and criticality of technical systems. Part 1 introduced the fundamental concept of unknown failures from an organizational, behavioral and cultural stand point. It also reported an industrial outlook to the problem, recent procedural standards, whilst discussing the financial implications and safety concerns. In this issue, the authors examine the technical aspects, reviewing the common causes of NFF failures in electronic, software and mechanical systems. This is followed by a survey on technological techniques actively being used to reduce the consequence of such instances. After discussing improvements in testability, the article identifies gaps in literature and points out the core areas that should be focused in the future. Special attention is paid to the recent trends on knowledge sharing and troubleshooting tools; with potential research on technical diagnosis being enumerated

    This paper presents the first part of a state of the art review on the No Fault Found (NFF) phenomenon. The aim has been to compile a systematic reference point for burgeoning NFF literature, and to provide a comprehensive overview for gaining an understanding of NFF knowledge and concepts. Increasing systems complexities have seen a rise in the number of unknown failures that are being reported during operational service. Units tagged as ‘NFF’ are evidence that a serviceable component was removed, and attempts to troubleshoot the root cause have been unsuccessful. There are many reasons on how these failures manifest themselves and these papers describe the prominent issues that have persisted across a variety of industrial applications and processes for decades. This article, in particular, deals with the impact of NFF from an organizational culture and human factors point of view. It also highlights recent developments in NFF standards, its financial implications and safety concerns

    The production of recombinant membrane proteins for structural and functional studies remains technically challenging due to low levels of expression and the inherent instability of many membrane proteins once solubilized in detergents. A protocol is described that combines ligation independent cloning of membrane proteins as GFP fusions with expression in Escherichia coli detected by GFP fluorescence. This enables the construction and expression screening of multiple membrane protein/variants to identify candidates suitable for further investment of time and effort. The GFP reporter is used in a primary screen of expression by visualizing GFP fluorescence following SDS polyacrylamide gel electrophoresis (SDS-PAGE). Membrane proteins that show both a high expression level with minimum degradation as indicated by the absence of free GFP, are selected for a secondary screen. These constructs are scaled and a total membrane fraction prepared and solubilized in four different detergents. Following ultracentrifugation to remove detergent-insoluble material, lysates are analyzed by fluorescence detection size exclusion chromatography (FSEC). Monitoring the size exclusion profile by GFP fluorescence provides information about the mono-dispersity and integrity of the membrane proteins in different detergents. Protein: detergent combinations that elute with a symmetrical peak with little or no free GFP and minimum aggregation are candidates for subsequent purification. Using the above methodology, the heterologous expression in E. coli of SED (shape, elongation, division, and sporulation) proteins from 47 different species of bacteria was analyzed. These proteins typically have ten transmembrane domains and are essential for cell division. The results show that the production of the SEDs orthologues in E. coli was highly variable with respect to the expression levels and integrity of the GFP fusion proteins. The experiment identified a subset for further investigation

    The cost of promiscuity: sexual transmission of Nosema microsporidian parasites in polyandrous honey bees

    Multiple mating (and insemination) by females with different males, polyandry, is widespread across animals, due to material and/or genetic benefits for females. It reaches particularly high levels in some social insects, in which queens can produce significantly fitter colonies by being polyandrous. It is therefore a paradox that two thirds of eusocial hymenopteran insects appear to be exclusively monandrous, in spite of the fitness benefits that polyandry could provide. One possible cost of polyandry could be sexually transmitted parasites, but evidence for these in social insects is extremely limited. Here we show that two different species of Nosema microsporidian parasites can transmit sexually in the honey bee Apis mellifera. Honey bee males that are infected by the parasite have Nosema spores in their semen, and queens artificially inseminated with either Nosema spores or the semen of Nosema-infected males became infected by the parasite. The emergent and more virulent N. ceranae achieved much higher rates of infection following insemination than did N. apis. The results provide the first quantitative evidence of a sexually transmitted disease (STD) in social insects, indicating that STDs may represent a potential cost of polyandry in social insects

    Bayesian paternity analysis and mating patterns in a parasitic nematode, Trichostrongylus tenuis

    Mating behaviour is a fundamental aspect of the evolutionary ecology of sexually reproducing species, but one that has been under-researched in parasitic nematodes. We analysed mating behaviour in the parasitic nematode Trichostrongylus tenuis by performing a paternity analysis in a population from a single red grouse host. Paternity of the 150 larval offspring of 25 mothers (sampled from one of the two host caeca) was assigned among 294 candidate fathers (sampled from both caeca). Each candidate father's probability of paternity of each offspring was estimated from 10-locus microsatellite genotypes. Seventy-six (51%) offspring were assigned a father with a probability of >0.8, and the estimated number of unsampled males was 136 (95% credible interval (CI) 77-219). The probability of a male from one caecum fathering an offspring in the other caecum was estimated as 0.024 (95% CI 0.003-0.077), indicating that the junction of the caeca is a strong barrier to dispersal. Levels of promiscuity (defined as the probability of two of an adult's offspring sharing only one parent) were high for both sexes. Variance in male reproductive success was moderately high, possibly because of a combination of random mating and high variance in post-copulatory reproductive success. These results provide the first data on individual mating behaviour among parasitic nematodes

    Costly mating delays drive female ornamentation in a capital breeder

    How fecundity might be traded off with mate attraction and other aspects of reproduction in females remains poorly understood. We investigated these allocation trade-offs using the common glowworm (Lampyris noctiluca), a lampyrid beetle, in which flightless, sedentary females only use resources gathered during the larval stage to attract flying males by glowing. While sexual signaling was not found to have a significant fecundity cost, a delay in successfully attracting a mate greatly increased the risk of reproductive failure, with fecundity losses being more severe in small females. These findings are among the first to show that failure to quickly attract a mate can decrease female fecundity. The results also show how the length of delay before mating can drive the evolution of female sexual ornamentation.Peer reviewe

    “Positive” Results Increase Down the Hierarchy of the Sciences

    The hypothesis of a Hierarchy of the Sciences with physical sciences at the top, social sciences at the bottom, and biological sciences in-between is nearly 200 years old. This order is intuitive and reflected in many features of academic life, but whether it reflects the “hardness” of scientific research—i.e., the extent to which research questions and results are determined by data and theories as opposed to non-cognitive factors—is controversial. This study analysed 2434 papers published in all disciplines and that declared to have tested a hypothesis. It was determined how many papers reported a “positive” (full or partial) or “negative” support for the tested hypothesis. If the hierarchy hypothesis is correct, then researchers in “softer” sciences should have fewer constraints to their conscious and unconscious biases, and therefore report more positive outcomes. Results confirmed the predictions at all levels considered: discipline, domain and methodology broadly defined. Controlling for observed differences between pure and applied disciplines, and between papers testing one or several hypotheses, the odds of reporting a positive result were around 5 times higher among papers in the disciplines of Psychology and Psychiatry and Economics and Business compared to Space Science, 2.3 times higher in the domain of social sciences compared to the physical sciences, and 3.4 times higher in studies applying behavioural and social methodologies on people compared to physical and chemical studies on non-biological material. In all comparisons, biological studies had intermediate values. These results suggest that the nature of hypotheses tested and the logical and methodological rigour employed to test them vary systematically across disciplines and fields, depending on the complexity of the subject matter and possibly other factors (e.g., a field's level of historical and/or intellectual development). On the other hand, these results support the scientific status of the social sciences against claims that they are completely subjective, by showing that, when they adopt a scientific approach to discovery, they differ from the natural sciences only by a matter of degree
